BACKGROUND: Spinal cord injury (SCI) is a devastating neurological disease characterized by neuroinflammation and neuronal apoptosis. The PI3K/AKT signaling pathway is related to the pathological process of SCI. Hematopoietic growth factor inducible neurokinin-1 type (HGFIN) is a transmembrane glycoprotein that exerts neuroprotective actions in various neurodegenerative diseases. However, the potential role and mechanism of HGFIN in the development of SCI are still unclear. OBJECTIVES: To investigate the effect of HGFIN on inflammation and neuronal apoptosis as well as the underlying mechanism in SCI. MATERIAL AND METHODS: A rat model of SCI was established, and Basso-Beattie-Bresnahan (BBB) motor function assay was performed to detect motor function. Expression of HGFIN was measured at 7 days after injury by western blot and immunofluorescence. An HGFIN-shRNA-carrying lentivirus was injected into the injury site to block the expression of HGFIN. The effects of HGFIN on neuronal apoptosis and the PI3K/AKT pathway were analyzed by TUNEL staining and immunofluorescence. The Iba-1 expression and the levels of pro-inflammatory cytokines were measured in spinal cord tissues by immunofluorescence staining and real-time polymerase chain reaction (PCR) analysis. RESULTS: The SCI rats showed increased expression of HGFIN in spinal cord tissues. The HGFIN deficiency aggravated SCI lesions, as evidenced by decreased BBB scores. At 7 days post-injury, HGFIN knockdown promoted neuronal apoptosis, accompanied by the increased expression level of the apoptosis effector cleaved caspase-3 and cleaved PARP, and decreased anti-apoptotic protein Bcl-2 expression. Moreover, HGFIN knockdown aggravated the inflammation process, indicated by increased Iba1-positive cells. The HGFIN knockdown increased the production of pro-inflammatory cytokines including IL-1ß, TNF-α and IL-6. Further analysis revealed that HGFIN deficiency reduced the activation of the PI3K/AKT pathway in spinal cord tissue after injury. CONCLUSIONS: Lentivirus-mediated downregulation of HGFIN exacerbates inflammation and neuronal apoptosis in SCI by regulating the PI3K/AKT pathway, and provides clues for developing novel therapeutic approaches and targets against SCI.

While climate models project that Greenland ice sheet (GrIS) melt will continue to accelerate with climate change, models exhibit limitations in capturing observed connections between GrIS melt and changes in high-latitude atmospheric circulation. Here we impose observed Arctic winds in a fully-coupled climate model with fixed anthropogenic forcing to quantify the influence of the rotational component of large-scale atmospheric circulation variability over the Arctic on the temperature field and the surface mass/energy balances through adiabatic processes. We show that recent changes involving mid-to-upper-tropospheric anticyclonic wind anomalies - linked with tropical forcing - explain half of the observed Greenland surface warming and ice loss acceleration since 1990, suggesting a pathway for large-scale winds to potentially enhance sea-level rise by ~0.2 mm/year per decade. We further reveal fingerprints of this observed teleconnection in paleo-reanalyses spanning the past 400 years, which heightens concern about model limitations to capture wind-driven adiabatic processes associated with GrIS melt.

Atmospheric rivers (ARs) that reach the complex terrain of High Mountain Asia (HMA) cause significant hydrological impacts for millions of people. While ARs are often associated with precipitation extremes and can cause floods and debris flows affecting populated communities, little is known about ARs that reach as far inland as HMA. This paper characterizes AR types and investigates dynamical mechanisms associated with the development of ARs that typically affect HMA. Combined empirical orthogonal function (cEOF) analysis using integrated water vapor transport (IVT) is applied to days where an AR reaches HMA. K-means cluster analysis applied to the first two principal components uncovered three subtypes of AR events with distinct synoptic characteristics during winter and spring months. The first subtype increases precipitation and IVT in Western HMA and is associated with a zonally oriented wave train propagating within the westerly jet waveguide. The second subtype is associated with enhanced southwesterly IVT, anomalous upper-level cyclonic circulation centered on 45 ∘ E, and precipitation in Northwestern HMA. The third subtype shows anomalous precipitation in Eastern HMA and southwesterly IVT across the Bay of Bengal. Interannual variations in the frequency of HMA ARs and relationships with various teleconnection patterns show that western HMA AR subtypes are sensitive to well-known remote large-scale climate factors, such as the El Niño Southern Oscillation, Arctic Oscillation, and the Siberian High. These results provide synoptic characterization of the three types of ARs that reach HMA and reveal the previously unexplored significance of their contribution to winter and spring precipitation. Consensus on the cause of recent midlatitude circulation changes toward a wavier manner in the Northern Hemisphere has not been reached, albeit a number of studies collectively suggest that this phenomenon is driven by global warming and associated Arctic amplification. Here, through a fingerprint analysis of various global simulations and a tropical heating-imposed experiment, we suggest that the suppression of tropical convection along the Inter Tropical Convergence Zone induced by sea surface temperature (SST) cooling trends over the tropical Eastern Pacific contributed to the increased summertime midlatitude waviness in the past 40 years through the generation of a Rossby-wave-train propagating within the jet waveguide and the reduced north-south temperature gradient. This perspective indicates less of an influence from the Arctic amplification on the observed mid-latitude wave amplification than what was previously estimated. This study also emphasizes the need to better predict the tropical Pacific SST variability in order to project the summer jet waviness and consequent weather extremes.

The observed upper (0-50 m) Arctic Ocean warming since 1979 has been primarily attributed to anthropogenically driven changes in the high latitudes. Here, using both observational and modeling analyses, we demonstrate that a multiyear trend in the summertime large-scale atmospheric circulation, which we ascribe to internal variability, has played an important role in upper ocean warming in summer and fall over the past four decades due to sea ice-albedo effect induced by atmospheric dynamics. Nudging experiments in which the wind fields are constrained toward the observed state support this mechanism and suggest that the internal variability contribution to recent upper Arctic Ocean warming accounts for up to one quarter of warming over the past four decades and up to 60% of warming from 2000 to 2018. This suggests that climate models need to replicate this important internal process in order to realistically simulate Arctic Ocean temperature variability and trends.

This work reports a facile and easily-achieved approach for enzyme immobilization by embedding glucose oxidase (GOx) in magnetic zeolitic imidazolate framework 8 (mZIF-8) via a de novo approach. As a demonstration of the power of such materials, the resulting GOx embedded mZIF-8 (mZIF-8@GOx) was utilized as a colorimetric sensor for rapid detection of glucose. This method was constructed on the basis of metal-organic frameworks (MOFs), which possessed very fascinating peroxidase-like properties, and the cascade reaction for the visual detection of glucose was combined into one step through the mZIF-8@GOx based mimic multi-enzyme system. After characterization by electron microscopy, X-ray diffraction, nitrogen sorption, fourier transform infrared spectroscopy and vibrating sample magnetometry, the as-prepared mZIF-8@GOx was confirmed with the robust core-shell structure, the monodisperse nanoparticle had an average diameter of about 200 nm and displayed superparamagnetism with a saturation magnetization value of 40.5 emu g(-1), it also exhibited a large surface area of 396.10 m(2) g(-1). As a peroxidase mimic, mZIF-8 was verified to be highly stable and of low cost, and showed a strong affinity towards H2O2. Meanwhile, the mZIF-8 embedded GOx also exhibited improved activity, stability and greatly enhanced selectivity in glucose detection. Moreover, the mZIF-8@GOx had excellent recyclability with high activity (88.7% residual activity after 12 times reuse).

Earth-abundant iron pyrite (FeS2) shows great potential as a light absorber for solar cells and photodetectors due to their high absorption coefficient (>10(5) cm(-1)). In this paper, high-quality phase-pure and single crystalline pyrite nanocrystals were synthesized via facile, low-cost, and environment friendly hydrothermal method. The molar ratio of sulphur to iron and the reaction time play a crucial role in determining the quality and morphology of FeS2 nanocrystals. X-ray diffraction and high-resolution transmission electron microscopy confirm that phase-pure and single crystalline pyrite nanocrystals can be synthesized with high sulphur to iron molar ratio and sufficient reaction time. For the first time, a crystalline nanogap pyrite photodetector with promising photocurrent and UV-visible photoresponse has been fabricated. This work further demonstrates a facile route to synthesize high-quality FeS2 nanomaterials and their potential in optoelectronic applications.

Rapid Arctic warming and sea-ice reduction in the Arctic Ocean are widely attributed to anthropogenic climate change. The Arctic warming exceeds the global average warming because of feedbacks that include sea-ice reduction and other dynamical and radiative feedbacks. We find that the most prominent annual mean surface and tropospheric warming in the Arctic since 1979 has occurred in northeastern Canada and Greenland. In this region, much of the year-to-year temperature variability is associated with the leading mode of large-scale circulation variability in the North Atlantic, namely, the North Atlantic Oscillation. Here we show that the recent warming in this region is strongly associated with a negative trend in the North Atlantic Oscillation, which is a response to anomalous Rossby wave-train activity originating in the tropical Pacific. Atmospheric model experiments forced by prescribed tropical sea surface temperatures simulate the observed circulation changes and associated tropospheric and surface warming over northeastern Canada and Greenland. Experiments from the Coupled Model Intercomparison Project Phase 5 (ref. 16) models with prescribed anthropogenic forcing show no similar circulation changes related to the North Atlantic Oscillation or associated tropospheric warming. This suggests that a substantial portion of recent warming in the northeastern Canada and Greenland sector of the Arctic arises from unforced natural variability.

Pine Island Glacier has thinned and accelerated over recent decades, significantly contributing to global sea-level rise. Increased oceanic melting of its ice shelf is thought to have triggered those changes. Observations and numerical modeling reveal large fluctuations in the ocean heat available in the adjacent bay and enhanced sensitivity of ice-shelf melting to water temperatures at intermediate depth, as a seabed ridge blocks the deepest and warmest waters from reaching the thickest ice. Oceanic melting decreased by 50% between January 2010 and 2012, with ocean conditions in 2012 partly attributable to atmospheric forcing associated with a strong La Niña event. Both atmospheric variability and local ice shelf and seabed geometry play fundamental roles in determining the response of the Antarctic Ice Sheet to climate.
